The victories of the newly promoted teams, 9 de Octubre and Chacaritas Jrs., marked the first round of the series B championship. Super-9 won 1-0 at home against Manta and the auriazules also won by the same score against Gualaceo.

The series and the round started at the Alejandro Ponce Noboa stadium in Guayaquil, where Gualberto Caicedo scored the winning goal for 9 de Octubre at the 25th minute, ending their 22-year absence from the top flight. Manta came close to equalizing, but Ángel Ledesma missed a penalty kick at the 49th minute. This match was played without spectators following the confirmation of a positive coronavirus case in Guayaquil by the Ministry of Health.
